This work focuses on the synthesis of comb-like materials where an anticancer drug, doxorubicin, is covalently linked with a hydrazone bond that allows the delivery of an higher amount of drug in acid environment. The hydrazone linkage was chosen also because when it is hydrolyzed it releases the intact doxorubicin structure. Three different kinds of materials are prepared in order to compare the changes in their properties: an amphiphilic biodegradable polymer drug where the doxorubicin is attached to poly(caprolactone) chains, a water soluble polymer-drug where the doxorubicin is attached to poly(ethylene glycol) chains, and a hydrophobic polymer-drug where the doxorubicin is attached to a poly(lactic acid) chain which is then used to produce nanoparticles. This work starts with the preparation of a comb-like polymer made up of hydrophilic and hydrophobic chains, PEGMA (poly(ethylene glycol) methacrylate) and HEMA-PCL (2-hydroxyethyl methacrylate poly(caprolactone)), respectively. This block copolymer can self-assemble to form small size micelles, that allow it to escape from renal exclusion, due to the high molecular weight and from reticuloendothelial system, due to the presence of PEG. The importance of the covalent bond is related to the possibility to avoid the burst release of the drug and thus to deliver doxorubicin for a long time, changing its biodistribution. In addition, the chosen acid-cleavable bond allows to control the release of doxorubicin, in other words, to improve the efficacy of the drug. These are advantages because in the physical entrapment, as doxorubicin is easily soluble in water, it is very difficult to deliver drug for long time. The same procedure is applied to a complete hydrophilic polymer, PEGMA OH-terminated in order to compare the final release properties of the obtained products. The produced materials were characterized by different analytical techniques, such as H-NMR, GPC, DLS, in order to verify their properties and their purity. The releases of doxorubicin from the polymers were investigated at different pH using several buffer solutions and they were monitored during the time. The results show a faster release of doxorubicin at lower pH.

Questo lavoro di tesi è basato sulla sintesi di materiali polimerici con struttura a pettine a cui è stata attaccata covalentemente, tramite legame idrazone, la doxorubicina, un farmaco antitumorale. L'utilizzo di una matrice polimerica con struttura ramificata offre il vantaggio di avere numerosi gruppi funzionali che possono essere opportunamente funzionalizzati in modo da poter attaccare un' elevata quantità di principio attivo. L'importanza del legame scelto, invece, risiede nella sensibilità che offre alla variazione del pH. In particolare, all'interno del corpo umano si registra un'alterazione dal valore di 7.4 nel flusso sanguigno a quello intorno a 5 nelle cellule tumorali. In questo modo, è possibile realizzare il rilascio controllato del farmaco nel tempo e nello spazio, in modo che solo le cellule malate vengano colpite. In altre parole, l'obiettivo è andare a migliorare l'efficacia del principio attivo usato. Nel corso di questo lavoro, sono stati sviluppati tre diversi tipi di materiali in modo da poterne confrontare le proprietà: un polimero biodegradabile anfifilico, in cui la doxorubicina è attaccata alle catene di poli(caprolattone), un polimero idrofilo dove il principio attivo è legato al poli(etilen glicole) e un polimero idrofobo, dove il farmaco è attaccato alle catene di acido polilattico. Questo ultimo materiale è stato usato per produrre nanoparticelle. Inizialmente, è stato prodotto il materiale anfifilico, costituito da PEGMA (poly(ethylene glycol) methacrylate) nella parte idrofila e da HEMA-PCL (2-hydroxyethyl methacrylate poly(caprolactone)) nella parte idrofoba. Il vantaggio risiede nel fatto che questo copolimero riesce a formare micelle di piccole dimensioni. Queste ne consentono la rapida eliminazione attraverso i reni, grazie al loro elevato peso molecolare, e dal sistema reticulo endoteliale, per la presenza del PEG. La stessa procedura è stata applicata per sintetizzare un polimero completamente idrofilo, in modo da confrontare i comportamenti dei due materiali. I prodotti ottenuti sono stati caratterizzati con varie tecniche analitiche in maniera tale da conoscerne le proprietà e avere conferma della loro struttura. Una volta ottenuti i materiali, sono stati monitorati nel tempo i rilasci della doxorubicina a diversi valori di pH, usando soluzioni tampone a pH noto. I risultati dimostrano che si verifica un rilascio più veloce a valori di pH più bassi.
