A novel microfluidic device to culture 3D cardiac microtissues was design, developed and validated. The new platform provides cells with mechanical deformation and ensures high resolution imaging and real-time monitoring of the cardiac electrical activity. Optimized protocol to generate healthy and pathological (i.e.,Duchenne muscular dystrophy) microtissues were presented and cardiac model organization and beating performances were investigated.
È stato progettato, sviluppato e validato un nuovo dispositivo microfluidico per la coltura in 3D di microtessuti cardiaci. La nuova piattaforma fornisce una deformazione meccanica alle cellule e permette l’ottenimento di immagini ad alta risoluzione e il monitoraggio in tempo reale dell’attività elettrica cardiaca. Sono stati sviluppati dei protocolli per generare microtessuti sani e patologici (distrofia muscolare di Duchenne) e l’organizzazione e il battito del microcostrutto sono stati analizzati.
