This Thesis work aim to deeply investigate how the novel strategy, which efficiency was already proved, combining the use of non-viral vectors and a vibration-based stimulation enhance transfection efficiency. In particular, the stimulation frequency was optimized for L929 cells transfected with PEI-based polyplexes in order to have the greater increasement of transfection efficiency. Thereafter, the effect of the stimulation was tested on different processes, such as cellular doubling time, endocytosis (my means of nanoparticles) and endosomal escape, to investigate which one was responsible of such transfection efficiency increasement.
Questo lavoro di Tesi ha l’obiettivo di investigare come una strategia di trasfezione innovativa, la cui efficacia è stata precedentemente verificata, combina l’uso di vettori non virali con una stimolazione di tipo vibrazionale per aumentare l’efficienza di trasfezione. In particolare, la frequenza di stimolazione è stata ottimizzata per le cellule L929 trasfettate con poliplessi a base di PEI affinché fosse possibile ottenere il maggior aumento dell’efficienza di trasfezione. Dopodiché, gli effetti della stimolazione sono stati testati su diversi processi, i quali il tempo di duplicazione delle cellule, l’endocitosi (usando delle nanoparticelle) e il rilascio dagli endosomi, per investigare quale fosse coinvolto nell’aumento dell’efficienza di trasfezione.
