The use of Drug Eluting Stents (DES) in non-vascular fields is, to date, very limited. In particular for the gastrointestinal tract, there is no currently approved DES for clinical practice. This thesis work, born from a collaboration with Dr. Lorenzo Dioscoridi, form Diagnostic and Interventional Digestive Endoscopy, Niguarda Ca' Granda Hospital, had the purpose of identifying and characterizing biodegradable polymers that can be used as coatings for gastrointestinal stents for the in situ controlled release of therapeutic agents. Materials were selected from the synthetic polymers approved by the Food and Drug Administration (FDA), to support a faster clinical application, and characterized to evaluate the release profiles and mechanical characteristics and to identify those with the characteristics suitable for the envisioned application.
L’applicazione dei Drug Eluting Stents (DES) in ambito non vascolare risulta ad oggi piuttosto limitata. In particolare per il tratto gastrointestinale, sul mercato mondiale attuale non c’è nessun DES approvato per l’utilizzo nella pratica clinica. Il presente lavoro di tesi, nato da una collaborazione con il dott. Lorenzo Dioscoridi, dell’Endoscopia Digestiva ed Interventistica dell'Ospedale Niguarda Ca’ Granda, ha avuto come scopo l’individuazione e la caratterizzazione di polimeri biodegradabili che possano essere utilizzati come rivestimenti di stent gastrointestinali per il rilascio localizzato di agenti terapeutici. I materiali sono stati selezionati tra i polimeri sintetici approvati dalla Food and Drug Administration (FDA), nell’ottica di favorire una più rapida applicazione clinica, e sono stati caratterizzati per valutarne i profili di rilascio e le caratteristiche meccaniche, individuando quelli con le proprietà idonee all’applicazione.
