This thesis aims at the formation of TiO2 nanotubes on Ti discs, in particular at the exploration of nanotubes with a bottleneck structure, exploiting voltage variations during the anodization, as a potential solution to tune the drug release from the surface of orthopedic implants. The work covers an analysis of the formation dynamics underlying the bottleneck structures, including how different parameters of time and voltage influence the morphology of the bottleneck nanotube, in particular variations of lengths and diameters. First, different voltage combinations were tested to find the optimal parameters, then the time variable was taken into consideration to obtain from one side a clear and ordered nanotubes morphology, on the other a greater reservoir capped with a short bottleneck. Moreover, an analysis of different drug loading techniques was performed, with ibuprofen used as a model drug. An efficient drug loading is in fact a key element for a correct release kinetics. At last, a preliminary study of ibuprofen release was performed comparing the performance of different nanotubular structures to bare Ti.
Questa tesi mira alla formazione di nanotubi di TiO2 su dischi di titanio, in particolare all'esplorazione di nanotubi con una struttura a collo di bottiglia, sfruttando le variazioni di tensione durante l'anodizzazione, come potenziale soluzione per regolare il rilascio di farmaci dalla superficie di impianti ortopedici. Il lavoro comprende un'analisi delle dinamiche di formazione alla base delle strutture a collo di bottiglia, includendo come diversi parametri di tempo e tensione influenzano la morfologia dei nanotubi a collo di bottiglia, in particolare le variazioni di lunghezza e diametro. In primo luogo, sono state testate diverse combinazioni di tensione per trovare i parametri ottimali, poi la variabile tempo è stata presa in considerazione per ottenere da un lato una morfologia chiara e ordinata dei nanotubi, dall'altro una struttura a serbatoio terminante con una strozzatura a collo di bottiglia. Inoltre, è stata effettuata un'analisi di diverse tecniche di caricamento del farmaco, con ibuprofene usato come farmaco modello. Un caricamento efficiente del farmaco è infatti un elemento chiave per una corretta cinetica di rilascio. Infine, è stato eseguito uno studio preliminare sul rilascio di ibuprofene dai campioni prodotti, confrontando le prestazioni di diverse strutture nanotubolari con quelle del titanio non trattato
