Coherent Raman scattering microscopy is a label-free, high sensitivity and non-invasive optical technique for analysing the chemical, morphological and structural composition of biological tissues, and can be used in the medical field for the diagnosis and classification of tumors. However, the measurement systems commonly used in research laboratories for this type of microscopy are not yet suitable for a clinical environment. In this context, a further need arises in order to exploit the potential of the technique: an engineered excitation laser that is compact, robust, portable and directly usable by medical personnel. An effective solution involves the use of a dual-cavity fiber laser source based on Single-Walled Carbon Nanotube (SWCNT) saturable absorber technology, which offers the dual benefit of achieving mode-locking and intrinsically synchronising the two output pulses. During my thesis work, I designed and built an experimental setup for the characterization of SWCNT saturable absorbers, I implemented the software to carry out the measurements and I measured the optical parameters of several samples. These were then inserted into the laser cavity and their operation was verified. In addition, I followed a measurement campaign on murine spines to show an example of how coherent Raman microscopy can be employed for pathology diagnosis, using a traditional system from which the engineered device originates.
La microscopia a diffusione Raman coerente è una tecnica ottica molto sensibile e non invasiva, che permette di analizzare la composizione chimica, morfologica e strutturale dei tessuti biologici senza l'utilizzo di marcatori esterni e può essere utilizzata in ambito medico per la diagnosi e la classificazione di tumori. Tuttavia, i sistemi di misura comunemente utilizzati nei laboratori di ricerca per questo tipo di microscopia non sono ancora idonei ad un ambiente clinico. Per poter sfruttare le potenzialità della tecnica, emerge quindi l'esigenza di un dispositivo laser ingegnerizzato, compatto, robusto, facilmente trasportabile e direttamente utilizzabile dal personale medico. Una soluzione efficace prevede l'utilizzo di una sorgente laser in fibra a doppia cavità basata sulla tecnologia degli assorbitori saturabili di tipo Single-Walled Carbon Nanotube (SWCNT), che offrono il duplice beneficio di raggiungere il regime di mode-locking e di sincronizzare intrinsecamente i due impulsi in uscita. Nel mio lavoro di tesi ho progettato e realizzato un setup sperimentale per la caratterizzazione degli assorbitori saturabili SWCNT, ho implementato il software per effettuare le misure e ho misurato i parametri ottici di diversi campioni che sono stati poi collocati all'interno della cavità laser, di cui è stato verificato il funzionamento. Ho inoltre seguito una campagna di misure su vertebre murine per mostrare un esempio di come la microscopia Raman coerente può essere utilizzata per la diagnosi di patologie, utilizzando un sistema tradizionale da cui ha origine il prodotto ingegnerizzato.
