Piattaforma microfluidica per lo studio dell'attivazione piastrinica nell'indagine della correlazione tra Forame Ovale Pervio ed emicrania

The Patent Foramen Ovale is a cardiac defect, found in about the 25% of the adult world population. Located at the interatrial septum, it consists in an opening that connects the two atria when the pressure within the right atrium exceeds the pressure of the left atrium. Research suggests a correlation between this conditions and disabling pathologies such as migraine and cryptogenic stoke, and several studies are investigating the possible factors that may explain this relationship, in order to identify the most suitable therapies. In this thesis, it has been considered the hypothesis that platelet activation induced by non- physiological shear stresses, generated within Patent Foramen Ovale, is the factor responsible for migraine symptoms. The aim of the project is to realize a microfluidic platform that reproduces those shear stresses in order to test their effect on platelet activation.

Il Forame Ovale Pervio è un difetto cardiaco presente nel 25% circa della popolazione adulta globale. Esso consiste in un’apertura presso il setto interatriale, che mette in comunicazione i due atri quando la pressione nell’atrio destro supera quella dell’atrio sinistro. Questa condizione sembra essere correlata a patologie invalidanti come emicrania e ictus criptogenici, e, per l’identificazione delle terapie più opportune, si stanno ricercando e studiando i fattori che possano spiegare questa relazione. In questo lavoro di tesi è stata considerata l’ipotesi che l’attivazione piastrinica indotta da sforzi di taglio non fisiologici, generati all’interno del Forame Ovale Pervio, sia il fattore responsabile dell’insorgenza dei sintomi emicranici. Lo scopo del progetto è di realizzare una piattaforma microfluidica in grado di riprodurre tali sforzi per testarne l’effetto sull’attivazione piastrinica.
