Time-Correlated Single Photon Counting (TCSPC) is a sensitive and precise optical detection technique widely used in fields such as biology, chemistry, and physics. It provides researchers with accurate measurements of the time delay between sample excitation and photon detection, revealing valuable insights into the sample's behaviour and structure. The TCSPC technique uses a pulsed laser to periodically stimulate a sample, measuring the time difference between the detected fluorescence photon and laser pulse. This data is then digitised and used to generate a histogram of photon-counts based on their arrival time. After a statistically significant number of repetitions, the produced histogram has the same shape as the light signal impinging on the photodetector. Although the typical TCSPC system relies on a single sensor and acquisition channel to generate a histogram based on photon arrival time, many applications require histograms that depend on other characteristics, such as spatial positioning of events. To achieve multi-dimensionality, additional acquisition channels are used in parallel. In this thesis, the first system presented was a 64-channel TCSPC system that required one of its boards to be adapted to perform timing measurements. The board developed for this thesis work not only facilitated timing measurements but also photon counting. As a result, this multi-dimensional system has an increased versatility in the range of applications for which it can be utilised. The second TCSPC system presented in this thesis is also multi-channel, it enables the parallelization of eight acquisition channels. This configuration accelerates measurements and enhances their precision using a Fast-TAC chip developed in the research laboratory. The board's FPGA firmware was developed in this thesis work and ensures the proper operation of this system.
Time-Correlated Single Photon Counting (TCSPC) è una tecnica di rilevamento ottico sensibile e precisa ampiamente utilizzata in campi come la biologia, la chimica e la fisica. Fornisce ai ricercatori misurazioni accurate del ritardo temporale tra l'eccitazione del campione e il rilevamento del fotone, generando importanti informazioni sul comportamento e sulla struttura del campione. La tecnica TCSPC utilizza un laser pulsato per stimolare periodicamente un campione, misurando la differenza di tempo tra il fotone di fluorescenza rilevato e l'impulso laser. Questi dati vengono quindi digitalizzati e utilizzati per generare un istogramma di conteggi di fotoni in base al loro tempo di arrivo. La scheda sviluppata in questo lavoro di tesi non solo ha facilitato le misurazioni temporali, ma anche il conteggio dei fotoni. Di conseguenza, questo sistema ha acquisito una maggiore versatilità nella gamma di applicazioni per le quali può essere utilizzato. Il secondo sistema TCSPC presentato in questa tesi è anch'esso multicanale, consente la parallelizzazione di otto canali di acquisizione. Questa configurazione non solo accelera le misurazioni ma anche ne migliora la precisione utilizzando un chip Fast-TAC sviluppato nel laboratorio di ricerca.
