Alzheimer’s disease (AD) is the most common cause of dementia nowadays. Despite the extensive research, the intricate interplay between the species involved continues to preclude a comprehensive understanding of the disease. The aim of this thesis is to analyse a new mathematical model that examines the evolution of the disease, focusing both on the amyloid-β protein and on key species involved in the pathogenesis of AD such as neurons, microglia and astroglia. Understanding the pathogenesis and the evolution of AD is crucial to capturing its fundamental dynamics. The study presents a new mathematical model based on the work of Puri and Li, coupling it with the Fisher-Kolmogorov equation to track also space dependence of the evolution of amyloid-β, the main component of the amyloid plaques found in the brains of people with AD. The aim is to study the temporal evolution of each species involved as well as their mutual interactions. First, we perform a study of the ODE model only, followed by a sensitivity analysis to measure the influence of individual coefficients. Secondly, we analyse the coupled Puri-Li and Fisher-Kolmogorov model on the graph representing each brain region as a node. Numerical simulations are carried out to approximate the evolution of each species across different brain regions. Within this context, a sensitivity analysis of the coefficients is performed as well to determine their impact on the temporal evolution of the species. Finally, a comprehensive study is carried out to investigate the influence of inter-regional interactions on disease progression.
L’Alzheimer è attualmente la causa più diffusa di demenza. I meccanismi complessi che stanno alla base della malattia non sono stati ancora del tutto compresi, nonostante le numerose ricerche nel corso degli anni. Lo scopo di questa tesi è analizzare un modello matematico che esamina la progressione temporale dell’Alzheimer, basato sulle interazioni tra neuroni, cellule della microglia, cellule dell’astroglia e amiloide-β. Per capire i meccanismi di interazione di queste specie è necessario uno studio approfondito della patogenesi e dell’evoluzione di questa malattia. La tesi sviluppa un modello non-lineare basato sul modello di Puri e Li, accoppiato con l’equazione di Fisher-Kolmogorov per studiare l’evoluzione della concentrazione di amiloide-β, la componente principale delle placche senili trovate nel cervello delle persone affette da Alzheimer, tenendo in considerazione anche la componente spaziale. Lo scopo di questa ricerca è trovare un modello che studia le progressioni temporali delle singole specie durante lo sviluppo della malattia e le loro interazioni. Una prima simulazione numerica per analizzare l’evoluzione temporale viene effettuata sul modello di ODE, seguita da un’analisi di sensibilità sui coefficienti presenti nel modello per studiare la loro influenza sulle concentrazioni. In seguito viene analizzato il modello accoppiato di Puri-Li e Fisher-Kolmogorov su un grafo in cui ogni nodo corrisponde a una specifica regione cerebrale. Anche in questo caso viene prima effettuata una simulazione numerica per vedere l’evoluzione temporale delle concentrazioni in ogni singola regione e, in seguito, un’analisi di sensibilità per vedere come le concentrazioni vengono influenzate dai coefficienti. Infine, un’ulteriore analisi di sensibilità permette di studiare l’influenza delle connessioni tra le regioni sulle concentrazioni.
