The aim of this thesis is to design a particle filter for coverslip mismatch and spherical aberrations estimation caused by the uncertainty of coverslip thickness in confocal microscopy. A model-based particle filtering approach has been developed in order to estimate the thick- ness mismatch and to retrieve the phase. Wiener models of the axial image concerning coverslip mismatch and spherical aberration are developed and used for the filtering problem. The particle filter is applied to solve the nonlinear filtering problem because of its high appli- cability in a general nonlinear non-Gaussian model. Its performance in terms of estimation error has been compared to the Extended Kalman Filter. The capabilities of the designed particle filters are shown both in simulation and by using a set of experimental data of two coverslips and one biological specimen. The mismatch and spherical aberration are estimated with a small error in simulation. In the experiment data, it is possible to find the best image correction, in which the aberration is minimum, in both the mismatch and spherical aberration estimation, although the spherical estimation shows a residual error.
In un microscopio confocale la causa principale di aberrazione sferica è dovuta dall’incertezza dello spessore del vetrino coprioggetto. L’obiettivo di questa tesi è implementare un filtro par- ticellare per la stima della discrepanza di spessore del vetrino coprioggetto e dell’aberrazione sferica. Un modello non lineare Wiener dell’immagine assiale è stato sviluppato per descrivere la discrepanza di spessore e l’aberrazione sferica e in seguito utilizzato per risolvere il problema di filtraggio. Il filtro particellare è utilizzato per stimare le grandezze di interesse data la sua versatilità e applicabilità in un generale modello non lineare e con rumori non Gaussiani. Le prestazioni del filtro particellare sono state confrontate con quelle del filtro di Kalman esteso, che si è dimostrato meno performante. La stima è stata condotta prima in simulazione e poi verificata con dati sperimentali considerando due vetrini copriogetto senza alcun campione da osservare e un vetrino con un campione biologico. In simulazione sia la discrepanza di spessore che l’aberrazione sferica sono state stimate correttamente. La stima condotta con dati sperimentali permette di conoscere correttamente il punto in cui l’aberrazione sferica è minima, sebbene la stima dell’aberrazione mostra dell’errore residuo.
