The basic idea of the Master thesis is the development of a Laser Scanning Microscopy for time-resolved Fluorescence measures. Using a couple of galvanometers, a laser light is deflected before entering in the microscope’s imaging system and then it’s focalized on the sample. The emitted fluorescence is collected and sent to a TCSPC system. In this way we obtain simultaneously spatial, spectral and time-resolved informations. On the other hand, the design and implementation of microscope system is accompanied with implementation of more efficient and less time-consuming data analysis algorithms concerning both spatial information and lifetime estimation.
L'idea principale alla base dell'esperienza di tesi è lo sviluppo di un microscopio a scansione laser per misure di fluorescenza risolte in tempo. Utilizzando una coppia di galvanometri, il fascio laser è deflesso prima di entrare nel sistema di imaging del microscopio e focalizzato sul campione. Il segnale di fluorescenza è inviato, tramite un fascio di fibre ottiche, ad una scheda TCSPC (Time-Correlated Single Photon Counting). In questo modo è possibile effettuare, conn un unico sistema, misure simultaneamente risolte in tempo, spettro e spazio. Inoltre sono stati sviluppati software di analisi delle misure di fluorescenza di fluorescenza più efficienti e veloci.
